大鼠生后发育期间胰岛D与A细胞相互关系的形态学观察

摘    要:①目的观察正常Wistar大鼠生后发育期间胰岛生长抑素细胞(D细胞)与胰高血糖素细胞(A细胞)间形态学关系,以探讨两者间功能调节的机理。②方法正常雄性Wistar大鼠20只,分为生后7,18,45d及成年组,每组5只。常规石蜡包埋,免疫组织化学PAP双重染色,分别显示大鼠胰岛D细胞与A细胞,光镜观察。③结果胰岛D细胞突起明显,与A细胞形成胞体-胞体、突起-胞体间的紧密接触,并可见D细胞呈新月形或其长胞突呈半环状及环状,部分或全部包绕A细胞。外分泌部也可见D细胞与A细胞接触紧密。生后7d与成年组相比D细胞与A细胞的相邻百分数具有显著性差异(F=4.06,q=4.75,P<0.05),其他各组间差异无显著性(q=0.65~3.62,P>0.05)。④结论D细胞可能通过旁分泌直接调节A细胞的功能活动,这种关系在发育期间不断增强

Abstract:Objective To investigate the relationship between pancreatic islet D cells and A cells of rat during postnatal development, and to discuss the possible regulation between these two kinds of cells. Methods Twenty male Wistar rats were evenly divided into 4 groups: 7th day after birth, 18th day after birth, 45th day after birth and adult rats. Pancreatic islet D cells and A cells were displayed with double staining of PAP immunohistochemical technique and observed under microscope. Results D cells had prominent processes, their cell bodies and processes formed close contact with A cells. Some crescent D cells or their ring like processes embraced A cells partially or completely. The close contact between D cells and A cells was also seen in the exocrine region. The contact percentage of D cells with A cells between 7th day after birth and adult group was significantly different( F=4.06,q=4.75,P <0. 05) . Conclusion Pancreatic islet D cells can regulate A cells by means of paracrine and this phenomenon is intensified gradually during postnatal development.
