盐酸异丙嗪对小鼠胚泡植入的影响

摘    要:目的 :研究盐酸异丙嗪对小鼠胚植入的影响。方法 :取孕 3~ 7d小鼠 ,每天上午 10时腹腔注射盐酸异丙嗪溶液 ;另取孕 4d小鼠 ,于上午 10时子宫角注入盐酸异丙嗪溶液 ,对照组同时用同法注射生理盐水 0 .2ml。于孕 8d检查胚胎植入数 ,同时 ,取子宫观察内膜的组织学及免疫组织化学变化。结果 :腹腔 2mg药物注射组抗植入率达 10 0 % ,腹腔 1mg药物组的抗植入率为 75 % ,子宫角药物注入组抗植入率达 75 % ,均明显少于对照组。组织学及免疫组化结果表明 ,盐酸异丙嗪可引起蜕膜细胞退化、解体 ,使蜕膜细胞不能正常形成纤维粘连蛋白 (FN)和层粘连蛋白 (LN)。结论 :盐酸异丙嗪对小鼠有明显的抗植入作用

Abstract:Objective: To evaluate the effect of promethazini hydrochloridum on mice blastocyst implantation .Methods: Pregnant mice were given promethazini hydrochloridum by intra-abdominal cavity injection every day from the 3rd day to 7th day of gestation and another group of mice were given the same drug by intra-uterus injection on 4th day of gestation.The numbers of embryo-implantation were examined and the histological and immunohistochemical variances in endometrium were observed. Results: The antiimplantation rates were 100% and 75% in the groups given 2mg promethazini hydrochloridum and lmg respectively treated throngh abdorminal cavity injection, and the antiimplantation rate was 75% in the group treated with 0.1mg drug throngh intra-uterus injection,showing a significant decrease than those of control groups given saline solution. Microscopic observation demonstrated that the decidua degenerated in the treated groups, and there was no positive staining of fibronection and laminin in the degenerated decidua. Conclusions: Promethazini hydrochloridum can produce significant effects on repressing mouse blastocyst implantation.
