乳腺癌PTEN、Akt基因蛋白的表达及临床意义

目的探讨prIEN在乳腺癌中表达与Akt、Cyclin D1、MMP-2蛋白表达的相关性及临床意义。方法采用链霉菌抗生物素蛋白-过氧化酶法（S-P法）检测PTEN、Akt、Cyclin D1、MMP-2蛋白在65例乳腺导管癌和15例正常乳腺和纤维腺瘤组织中的表达。结果15例正常乳腺和纤维腺瘤组织中PTEN表达最高。65例乳腺导管癌中，淋巴转移32例，无淋巴转移33例，PTEN的表达率在乳腺导管原位癌和分化较好（Ⅰ、Ⅱ）的乳腺浸润性导管癌中表达率高，与Ⅲ、Ⅳ期乳腺浸润导管癌中PTEN表达有明显差异（P〈0．01）。PTEN表达与Akt、Cyclin D1、MMP-2表达呈负相关。结论PTEN蛋白表达下降发生在乳腺癌的早期，并且与乳腺癌的发展预后密切相关。PTEN蛋白减少促使P13K／Akt信号转导通路的活性可能是乳腺癌发生发展的分子机制，Cyclin D1、MMP-2参与了该过程。

Objective to investigate the relativity and clinic significance of expression of protein PTEN and expression of protein Akt,Cyclin D1,MMP-2 in breast cancer.
