| 摘 要： | 目的用RNA i方法抑制胰腺癌细胞PANC-1中凋亡抑制蛋白survivin的表达。方法用免疫细胞化学技术和RT-PCR检测胰腺癌细胞PANC-1中凋亡抑制蛋白survivin的表达,构建针对survivin基因的siRNA表达载体并进行酶切和测序鉴定,用脂质体转染胰腺癌细胞PANC-1,RT-PCR初步分析siRNA表达载体对survivin表达的抑制作用。结果survivin在胰腺癌细胞PANC-1中有较高表达,siRNA表达载体可以有效的抑制survivin的表达,抑制率达60%。结论用RNAi表达载体可以有效抑制胰腺癌细胞PANC-1中凋亡抑制蛋白survivin的表达。

| Abstract: | Objective To inhibit the expression of survivin in pancreatic cancer cell PANC-1 with RNAi. Methods The expression of survivin was examined with RT-PCR and immunocytochemistry, and the siRNA expression vectors targeting survivin were constructed and transfected into PANC-1 cells with liposomes. The expression of survivin mRNA was detected with RT-PCR. Results There was a high degree expression of survivin in PANC-1 cells; the expression of survivin was inhibited by about 60% by the siRNA expression vectors. Conclusion The siRNA expression vector could inhibit the expression of survivin in PANC-1 cells effectively. |
