应用CD45/SSC设门的流式细胞术对骨髓增生异常综合征免疫分型

目的:探讨骨髓增生异常综合征(MDS)患者的免疫表型特片。方法:对54例 MDS 患者的骨髓细胞悬液应用三色流式细胞术和一组单克隆抗体对 CD45/SSC 设门提取的原始细胞进行免疫表型分析。结果:54例 MDS中,原始细胞阳性表达 CD34、CD13、CD33、CD2、CD7、CD10和 CD19的患者分别为27例(50.0%)、43例(79.6%)、39例(72.2%)、8例(14.8%)、18例(33.3%)、4例(7.4%)和3例(5.5%);9/9例(100%)患者呈 HLA-DR 阳性,3/9例(33.3%)患者CD117阳性,5/47例(10.6%)患者 CD14阳性,2/8例(25.0%)患者 CD15阳性,1/14例(7.1%)患者 CD3阳性.2/37例(5.4%)患者 CD20阳性,15/15例(100%)患者 CD22均阴性。难治性贫血/伴环形铁粒幼细胞的难治性贫血(RA/RARS)、伴原始细胞增多的难治性贫血(RAEB)和转变中的 RAEB(RAEB-T)3组间各免疫表型差异无显著性。结论:免疫分型是 MDS 诊断中对细胞形态学和细胞遗传学检查的重要补充。

Immunophenotypic features of myelodysplastic syndromes by CD45/SSC gating and flow cytometry

Objective To explore the immunophenotypic features of blasts in the patients with myelodysplastic syndromes (MDS).Methods A panel of monoclonal antibodies and flow cytometry using CD45/SSC gating strategies were used for the immunophenotype in the bone marrow nucleated cells of 54 patients with MDS.Results Of 54 MDS patients,the blasts of 27(50.0%),43(79.6%),39(72.2%),8(14.8%),18(33.3%),4(7.4%) and 3(5.5%) patients had positive expressions of CD34.CD13,CD33,CD2,CD7,CD10 and CD19,separately.There were 100%(9/9),33.3%(3/9),10.6%(5/ 47),25.0%(2/8),7.1%(1/14),5.4%(2/37),and 0%(0/15) of MDS pateints who were positive for HLA-DR,CD117,CD14, CD15,CD3,CD20 and CD22,respectively.However,there was no significant difference in the expression of the above- mentioned antigens on the blasts in the bone marrow between the patients with different MDS subtypes.Conclusions The immunophenotype is an important complementarity to the morphology and cytogenetics for the diagnosis of MDS.
