接骨丸对SD大鼠肝肾毒性的实验研究

摘    要:目的:观察接骨丸对SD大鼠肝脏和肾脏的毒性,完善接骨丸质量标准,确保该制剂使用安全。方法:选择40只健康SD大鼠,雌雄各半,按照数字随机表法分为空白对照组、接骨丸低剂量组、接骨丸高剂量组及接骨丸生马钱子组,每组10只。实验开始前取每只大鼠的血液测定肝肾功能相关的生化指标,包括丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、碱性磷酸酶(ALP)、总胆红素(T-BIL)、血糖(GLU)、尿素氮(BUN)、肌酐(Cr)及三酰甘油(TG)。对每组SD大鼠按照实验设定的给药剂量连续12周灌胃,每日1次,每周测定大鼠体质量,并根据体质量实时调整给药量。给药期间每日观察两次大鼠的活动及行为,记录变化情况。12周灌胃结束后,取每只大鼠的血样测量实验前测量的生化指标并比较。停药2周后再次进行上述指标检测并比较,处死小鼠取小鼠肝脏、肾脏测量肝、肾脏器系数,取肝组织和肾组织制片观察大鼠的病理改变。结果:40只小鼠在实验期间的活动及行为表现均无明显异常;实验前4组小鼠的血生化指标无明显差异;灌胃12周后、停药2周后,空白组、低剂量组小鼠的血生化指标和实验前比较无明显变化,灌胃12周后,高剂量组和生马钱子组小鼠的ALT、AST、ALP、TBIL、GLU、BUN、Cr及TG均较实验前有明显上升,前后比较差异具有统计学意义(P〈0.05),生马钱子组小鼠上述指标的上升幅度明显高于高剂量组,差异具有统计学意义(P〈0.05)。停药2周后,高剂量组和生马钱子组小鼠的上述生化指标均得到明显下降,但与实验前比较,差异有统计学意义(P〈0.05)。停药2周后空白组、低剂量组小鼠的肝、肾脏器系数处于正常范围,而高剂量组和生马钱子组小鼠的肝肾脏器系数明显超出正常范围,差异有统计学意义(P〈0.05)。结论:按照质量标准生产的接骨丸在

Abstract:Objective:To study bone pill on the SD rat liver and kidney toxicity,improve bone pill quality standards to ensure safe use of this preparation. Methods :40 healthy male and female SD rats, according to a random number table method will be divided into four groups( control group,low dose group and high dose group and bone health Strychnos pill group) , n = 10 only. Before the experiment began to take each rat blood measuring biochemical markers of liver and kidney function related (alanine aminotransferase (ALT), aspartate aminotransferase ( AST), alkaline phosphatase ( ALP), total cholesterol red pigment ( T-BIL), glucose ( GLU ), blood urea nitrogen (BUN) ,creatinine(Cr) and triglyceride(TG). SD rats for each group according to the dose of experimental set garage for 12 weeks, once daily, body weight was measured every week, and the dosage adjusted in real time according to the weight of administration twice daily observation during the activities and behavior in rats, record changes, administered 12 weeks after the end of each take blood samples were measured in rats before the experiment and compare the biochemical measurements, withdrawal two weeks after the test again and compare these indicators, the mice were sacrificed mice were liver, kidney measuring liver, kidneys coefficients, liver tissue and kidney tissue system piece pathological changes observed in rats. results :40 mice during the experiment activities and behavior no obvious abnormalities;before the experiment four groups of mice showed no significant difference in blood biochemistry; administered 12 weeks after discontinuation of 2 weeks later, the control group,lowdose group of the blood biochemical parameters and no significant change before the experiment,by gavage for 12 weeks,the high-dose group and the Health and brucine mice ALT, AST, ALP,T-BIL, GLU, BUN,Cr,and TG increased significantly compared with before the experiment, a statistically significant difference between before and after( P ?
