摘    要:目的:研究人乳腺癌组织转移相关基因表达谱,观察乳腺癌转移过程中有关基因的功能。方法:挑选399个与肿瘤转移相关的基因克隆,经PCR纯化后,点布于多聚赖氨酸包备的玻片上。提取正常乳腺组织及乳腺癌组织的总RNA,反转录后标记cDNA探针,与cDNA微阵列杂交。使用ScanArray4000共聚焦激光扫描仪扫描芯片。结果:81个基因与乳腺癌组织转移相关,包括癌基因、抑癌基因、运动因子、粘附因子、细胞凋亡调节因子、基质金属蛋白酶、血管形成因子等。结论:多个基因参与乳腺癌转移过程,它们共同作用,调节肿瘤细胞的生物学特性,促使乳腺癌转移和进展。

Abstract:Objective: To understand the molecular mechanism of metastasis of breast cancer. Method: The PCR porducts of 399 genes were spotted on to the slides coated with poly-lysine. Total RNA from human breast cancer and normal tissue were extracted and labelled by fluorescent cy-dye. The labelled probes were hybridized to the cDNA microarray and scanned with ScanArray 4000. Rusults: Specific mRNA expression profiles related to the metastasis of breast cancer were got. There were many genes envolved in the metastasis of breast cancer, including oncogenes, tumor-suppressor genes, motify factors, adhension moleculars, apoptosis genes, extrcellular metrix enzymes, vascularization factors, etc. Conclusion: Many genes are envolved in the process of the metastasis of breast cancer, interacted or interfered with each other, and influence the biological properties of tumor cells which eventually make the metastasis of tumur happen.
