汉黄芩素对胶质瘤U251细胞增殖、凋亡、Survivin及Caspase-3表达的影响

摘    要:目的探讨汉黄芩素在人胶质瘤患者中对U251细胞的增殖与凋亡及对存活素(Survivin)及半胱天冬酶-3(Caspase-3)表达的影响。方法不同浓度汉黄芩素作用于人胶质瘤96株U251细胞,并与使用替莫唑胺(TMZ)的阳性对照组(48株)及未给予任何药物的空白对照组(48株)对比细胞增殖抑制率、细胞凋亡率及Survivin和Caspase-3蛋白的表达强度。结果各浓度汉黄芩素对细胞增殖抑制率均显著高于空白对照组,且随药物浓度及作用时间的增加呈现出显著增高趋势(P〈0.05),汉黄芩素组细胞凋亡率显著高于TMZ组(P〈0.01),TMZ组显著高于空白对照组(P〈0.01),在汉黄芩素组内,细胞凋亡率随着药物浓度增加显著增加(P〈0.05);汉黄芩素组Survivin表达强度佩著低于TMZ及空白对照组(P〈0.01);两药物组Caspase-3表达强度显著高于空白对照组(P〈0.01)。结论汉黄芩素可通过调节Survivin、Caspase-3蛋白的表达对胶质瘤U251细胞起到诱导凋产的作用。

Abstract:Objective To discuss the effects of wogonin on proliferation, apoptosis, Survivin, and Caspase-3 expression in human giloma cell line U251. Methods Ninety-six human glioma U251 cells were cultured in different concentration of wogonin, and the inhibitions of cell proliferation, apoptosis rates, Survivin and Caspase-3 expression intensity were contrasted between the groups. Results The cell proliferation inhibition rate in each group with different concentration of wogonin was significantly higher than the blank control group, and the cell proliferation inhibition rate increased significantly with the increase of drug concentration and time (P 〈 0.05); the cell apoptosis rates in wogonin group were significantly higher than TMZ group (P 〈 0.01), and the TMZ group significantly higher than the blank control group (P 〈 0.01), and the cell apoptosis rates increased significantly with the increase of drug concentration in wogonin group (P 〈 0.05); Survivin expression intensity in wogonin group were significantly lower than the other groups (P 〈 O.O1), and Caspase-3 expression intensity in blank control group were significantly significantly lower than the other groups (P〈 0.01). Conclusion Wogonin plays a role in inducing apoptosis in glioma U251 cells through regulation of Survivin, Caspase-3 protein expression.
