人胎盘间充质干细胞大鼠体内移植的初步免疫学研究

摘    要:目的了解大鼠对于人胎盘间充质干细胞是否存在天然免疫耐受,分析人胎盘间充质干细胞在大鼠体内出现免疫排斥的可能。方法分离提取并鉴定人胎盘间充质干细胞,观察大鼠尾静脉移植人胎盘间充质干细胞后有无出现相关的免疫排斥症状、细胞在大鼠体内的分布情况及脏器的病理改变。结果大鼠尾静脉移植人胎盘间充质干细胞后未出现急慢性免疫排斥反应,肝、肾、肺可见标记后的人胎盘间充质干细胞的分布,但相关病理切片未见明显的淋巴细胞浸润和纤维组织增生现象。结论人胎盘间充质干细胞与大鼠脏器间存在天然的免疫耐受。

Abstract:Objective To understand the distribution of human placenta-derived mesenchymal stem cells (HPDMSCs) in rats by intravenous injection, and to evaluate the possibility of immune rejection in rats as a result of transplantation of HPDMSCs. Methods After isolating MSCs from the human placenta, the immunophenotypes of cell was observed by flow cytometry, and the rats received an injection of PKH26-1abeled HPDMSCs via the caudal vein. After the success- ful transplantations,the distribution of HPDMSCs in rats was detected by fluorescence microscope, and the organs conditions of the rats were also observed. Results The results showed that intravenous injection had no obvious acute and chronic immune rejection.In addition, HPDMSCs was detected in the liver,kidney cortex,lung,but the pathology of related organs had no obvious lymphocyte infiltration and proliferation of fibrous tissue. Conclusion Natural im- mune tolerance may exist between HPDMSCs and rat organs.
