目的:探讨乙酰肝素酶(HPA)反义寡脱氧核苷酸(AS-OND)对人胃癌细胞株SGC-7901细胞基质金属蛋白酶-2(MMP-2)及组织金属蛋白酶抑制剂-2(TIMP-2)表达的影响。方法:采用脂质体介导法将HPAAS-ODN转染SGC-7901细胞G用RT-PCR检测HPA-mRNA表达,用免疫细胞化学的方法检测细胞内MMP-2和TIMP-2的变化。结果:AS-ODN转染48h后GSGC-7901细胞内HPA-mRNA表达下降(P<0.01),MMP-2的表达量亦减少(P<0.01)。结论:HPAAS-ODN可以有效地抑制胃癌细胞MMP-2的表达。

Influence of MMP-2 and TIMP-2 expressions in SGC-7901 by heparanase antisense oligodeoxynucleotide

Objective: To evaluate the effect of heparanase antisense oligodeoxynucleotide(AS-OND) on the expression of matrix metalloproteinase-2 (MMP-2)and tissue inhibitor of metalloproteinase-2 (TIMP-2)in human gastric carcinoma cell line SGC-7901. Methods: Human gastric adenocarcinoma cell line SGC-7901 was transfected with liposome-mediated heparanase AS-OND. The expression of heparanase mRNA was detected after transfection by semi-quantitative total RNAs RT-PCR. The changes of MMP-2 and TIMP-2 were observed by immunocytochemistry. Results: The expressions of heparanase mRNA and MMP-2 of SGC-7901 cells treated with AS-ODN of different final concentrations were significantly decreased 48 hours later compared with those of the controls (P<0.01). Conclusion: Heparanase AS-ODN has a significant inhibitory effect on the expression of MMP-2 in human gastric carcinoma cell line SGC-7901 .
