骨髓基质干细胞移植治疗大鼠脊髓挫伤对脊髓脑源性神经生长因子表达的影响

摘    要:目的:探讨骨髓基质干细胞移植治疗脊髓挫伤后脊髓腹角早期脑源性神经营养因子(brain-derived neurotrophic factor,BDNF)的表达变化。方法:转绿色荧光蛋白基因骨髓基质干细胞移植治疗大鼠脊髓挫伤,脊髓冰冻切片行BDNF免疫组化ABC染色。结果:各移植组均见绿色荧光物质。脊髓挫伤后1dBDNF阳性细胞开始增多,7d达高峰,21d仍有较高表达。同一时间段内与挫伤对照组相比,移植组阳性细胞均增多,1d、3d和21d有显著性差异;3d和14d移植组平均灰度值有显著性差异;1d和14d移植组MOD有显著性差异。结论:脊髓挫伤后移植骨髓基质干细胞可以到达脊髓损伤处并可增加挫伤脊髓BDNF表达。

Abstract:Objective: To explore the change of brain-derived neurotrophic factor (BDNF) expression of the contused spinal cord in the early stage following transplantation of bone marrows stromal stem cells (BMSCs) in rats.Methods: The modified method of Allen was employed to establish contused spinal cord injury models. After the contused spinal cord, GFP-BMSCs were immediately transplanted into rats via caudal vein. The frozen sections were observed under fluorescent microscope and the distribution and expression of BDNF in the ventral horn of the spinal cord was detected by immunohistochemical ABC staining.Results: Green fluoroscent cycle dots were found in all BMSCs transplantation groups. BDNF expression began increasing following administration, reached peak after 7d and kept high expression level after 21d. During the same period, BDNF positive expression in BMSCs transplantation group was higher than that in the control group. There was significant difference in 1d, 3d and 21d groups, and there was significant difference of the mean gray level between 3d and 14d groups compared with the control group.Conclusion: The transplanted BMSCs after the contused spinal cord can significantly increase the expression of BDNF in the spinal cord.
