靛玉红对溃疡性结肠炎小鼠CD4CD25Treg细胞Foxp3表达的影响

目的观察青黛及其有效成分靛玉红对溃疡性结肠炎（UC）小鼠脾脏CD4CD25Treg细胞Foxp3表达的影响，探讨其治疗uc的作用机制和靶点。方法将50只BABL／c小鼠随机分为正常组、模型组、靛玉红组、青黛组和5-ASA组。除正常组外，其余小鼠采用自由饮用5％葡聚糖硫酸钠（DSS）溶液5天，制备DSS诱导的UC小鼠模型。于造模后第5天开始，予各组小鼠给药治疗7天。观察各组小鼠一般状况及结肠组织病理学改变，同时检测脾脏CD4CD25Treg细胞Foxp3的表达变化。结果与正常组比较，模型组小鼠CD4CD25Treg细胞Foxp3的表达显著减少（P〈0．01）；各治疗组CD4CD25Treg细胞Foxp3的表达较模型组升高（P〈0．01）。结论青黛及其有效成分靛玉红可通过上调CD4CD25Treg细胞Foxp3表达以防治UC。

Influence of Indirubin on Foxp3 expression of CD4CD25 regulatory T cells in ulcerative colitis mice

Objective To observe the effects of Qingdai（ Indigo Naturalis） and Indirubin, an active ingredient of Indigo Naturalis on the expression of Foxp3, a key marker of CD4CD25 regulatory T cells, in the mice ulcerative colitis（ UC）, in an attempt to study their action mechanisms and targets. Methods Fifty BABL/c nice were randomized into five groups： normal group, model group, Indirubin group, Qingdai group and 5-ASA Group. The mice except for those in the normal group were given 5% dextran sulfate sodium（DSS） for 5 days to establish UC model. From the fifth day, corresponding treatments were applied to these groups for total seven days. The general conditions, the histological appearance of the colon and the change of Foxp3 expression were observed. Results Significant reduction of Foxp3 expression was found in the model group compared with the normal group （ P 〈 0.01 ）, and Foxp3 expressions were significantly increased in all treat- ment groups while compared with the model group （ P 〈 0.01 ）. Conclusion Qingdai （ Indigo Naturalis） and Indirubin can upregulate expression of Foxp3 of CD4CD25 regulatory T ceils to manage ulcerative colitis.
