P27在急性白血病患者中表达的研究

目的 :研究细胞周期蛋白依赖激酶抑制物P2 7在急性白血病 (AL)患者中的表达 ,探讨P2 7在AL中的作用机制及其临床意义。方法 :应用免疫细胞化学链霉素抗生物素蛋白 过氧化酶 (SP)法检测AL患者 4 3例及对照组 10例P2 7的表达 ,并用蛋白印迹 (Western blot) DAB(3,3’二氨基联苯胺 )显色与化学发光显示 ]法检测其中 11例AL患者及 10例对照P2 7的表达。结果 :SP法显示 4 3例AL患者P2 7阳性表达率 (18.6 0 % )明显低于对照组 (80 % ) (P <0 .0 5 )。急性淋巴细胞白血病 (ALL)患者阳性表达率 (2 2 .73% ) ,急性非淋巴细胞白血病(ANLL)患者 (14 .2 9% )均低于对照组 (均P <0 .0 5 )。但两者间差异无统计学意义 (P >0 .0 5 ) ;未缓解组P2 7阳性表达率较缓解组患者低 ,但两者间差异无统计学意义。蛋白印迹法也同样提示AL患者P2 7呈低表达。结论 :P2 7在AL患者中有异常低表达 ,但未显示与缓解相关性。P2 7在AL中是否具有抑癌基因的作用 ,有待进一步研究。

The study of expression of P27 in acute leukemia

Objective:To study expression of P27, its function and clinical value in patients with acute leukemia(AL).Method:The expression of P27 was measured in the bone marrow samples of 43 AL and 10 cases in control group by S-P immunocytochemical method, 11 AL and 10 cases in control group were examined by western-blot DAB method and chemical luminescene.Result:The positive rate of P27 in 43 AL was 18.60% , which was apparently lower than that in the control group with the rate of 80%(P< 0.05). The expression of P27 in patients with ALL was 14.29% and 22.73% in patients with ANLL, which were lower than that of the control, however, had no significant difference between them (P> 0.05). There also had no significant difference between the remission group and the non-remission group. Similiar result could been seen by western-blot method.Conclusion:P27 had lowexpression in AL but was not related to remission. Whether P27 in AL be an anti-oncogene need further to study.
