抗氧化微量营养素对T1DM小鼠胰岛细胞保护作用的超微结构研究

摘    要:目的探讨联合抗氧化微量营养素(antioxidant micronutrients,AM)对胰岛β细胞超微结构的保护作用。方法应用多低剂量链脲佐菌素法(multipe low dosage of streptozotocin,MLDS)方法制备T1DM(type 1 diabetes mellitus)小鼠模型,分别联合添加4种AM[硒(Se)+维生素E(VE)+钒(V)+铬(Cr)]和7种AM(Se+VE+V+Cr+VC+硫辛酸+烟酰胺)进行干预,观察胰岛β细胞超微结构的变化。结果联合补充AM使T1DM小鼠血糖明显降低;T1DM模型组胰岛β细胞及内分泌颗粒数量减少,内质网明显扩张囊泡变,部分线粒体发生肿胀变性;AM4、AM7组小鼠胰岛β细胞超微结构完好,胞浆内胰岛素分泌颗粒数量明显增多于T1DM模型组小鼠。结论联合AM对T1DM模型小鼠胰岛β细胞超微结构具有良好的保护作用。

Abstract:Objective To observe the effects of antioxidant micronutrients(AM) on β cells ultra-structure in islets of type one diabetic mellitus(T1DM) mice.Method T1DM mice model was made by injection of multiple low dosages of streptozotocin(MLDS).The supplementation of AM was designed two groups:AM4 selenium(Se)+VE+vanadium(V)+chromium(Cr) and AM7(Se+VE+V+Cr+VC+lipoic acid+ niacinamide).The ultrastructure observation of islets in T1DM mice was finished by electron microscope.Results There were decrease of β cells with endocrine granules,degeneration of endoplasmic reticulum and mitochondria,and proliferation of α cells of islets in T1DM model group.However,theβcells with endocrine granules were abundant and the ultra-structure of islets were normal in both AM4 and AM7 groups.Simultaneously,the blood glucose level of mice in AM4 and AM7 groups significantly decreased compared with that of T1DM model group.Conclusion Antioxidant micronutrients may protect ultra-structure of islets βcells against oxidative destruction in T1DM.
