核黄素对异丙肾上腺素所致的缺血后再灌注损伤大鼠心肌的保护作用

摘    要:目的:观察核黄素对异丙肾上腺素(isoproterenol,ISO)引起的心肌缺血后的离体心肌缺血再灌注损伤大鼠心肌的保护作用。方法:健康雄性SD大鼠30只,随机分为3组,即正常对照组(NC),心肌缺血组(MI),核黄素预处理组(RBF),每组10只。采用大剂量ISO复制大鼠心肌缺血损伤模型,测定3组血清中过氧化氢酶(CAT)活性,乳酸脱氢酶(LDH)含量及血小板聚集性;随后3组均采用Langendroff离体心脏灌流装置再复制心肌缺血再灌注模型,分别测定心肌匀浆中CAT活性,LDH及丙二醛(MDA)含量。结果:与MI组相比较RBF组可明显降低血清中LDH含量(P<0.01)并可明显抑制血小板聚集性(P<0.05),显著提高血清CAT活性(P<0.05);离体心脏缺血再灌注后RBF组可显著提高MI组心肌匀浆中的CAT活性(P<0.05),显著降低MDA含量(P<0.05)。结论:核黄素对ISO所致的心肌缺血后的离体心肌缺血再灌注损伤有明显的保护作用,其机制可能与抑制脂质过氧化,稳定细胞膜和改善微循环有关。

Abstract:Objective:To observe the protective effect of riboflavin on ischemia-reperfusion injury followed by myocardial ischemia induced by isoproterenol(ISO) in rats.Methods: 30 fasted male rats were randomly divided into three groups: normal control group(NC),myocardial ischemia group(MI) and riboflavin group(RBF).Myocardial ischemia was induced by subcutaneous injection of ISO,and LDH and CAT in serum and platelet aggregation were measured.The model of myocardial ischemia-reperfusion set up by Langendroff device was used to observe the changes of the content of LDH,CAT and MDA in the myocardial homogenate.Results: The results showed that riboflavin could significantly reduce the content of LDH in serum(P<0.01)and platelet aggregation(P<0.05)and it could increase both the activity of CAT in serum(P<0.05)and the activity of CAT(P<0.05),but reduced the content of MDA(P<0.05) in the myocardial homogenate after ex vivo myocardial ischemia-reperfusion compared with MI group.Conclusion: Riboflavin plays a protective role in ischemia and ischemia-reperfusion injury myocardium in rats,which is related to increasing the activity of enzyme that can scavenge oxygen free radicals and protect the cells by meliorating microcirculation.
