摘    要:目的通过采用高效液相色谱法测定不同加工方法所得金银花药材中绿原酸和木犀草苷的含量,考察不同加工方法对金银花质量的影响。方法绿原酸含量测定采用AgilentZorbax SB—C18色谱柱(4.6mm×250mm,5um),流动相:乙腈-0.4%磷酸(15:85);流速:1.0mL·min-1;检测波长:327nm;柱温:室温。木犀草苷含量测定采用Hypersil Pheny1-2色谱柱(4.6mm×150mm,5Ixm),流动相:乙腈一0.5%冰醋酸,二元梯度洗脱,0~15min,乙腈10%~20%,15~30min,乙腈20%,30—40min,乙腈20%-30%;流速:1.0mL·min~;检测波长:350nm;柱温:40℃。结果不同加工方法对金银花药材中绿原酸和木犀草苷的含量有一定影响。结论烘干法和微波法所得样品质量较好,且药材的外观色泽与药材内在质量之间存在较为密切的关系。

Determination of chlorogenic acid and galuteolin With different processing methods in flos lonicerae by HpLC

Abstract:Objective To investigate the effect of different processing methods on the content of chlorogenic acid and galuteolin inflos lonicerae and the quality offlos lonicerae. Methods The determination of chlorogenic acid was performed on Agilent Zorbax SB - C18 column (4.6 mm × 250 mm,5 um ). The mobile phase was acetonitrile - 0.4% phosphate ( 15 :85) with the flow rate of 1.0 mL · min-1. The detection wavelength was 327 nm and the column temperature was room temperature. The determination of galuteolin was performed on Hypersil Phenyl -2 column(4. 6 mm ×150 mm,5 um). The mobile phase was acetonitrile -0. 5% glacial acetic acid with binary gradient elution, acetonitrile 10% - 20% within 0 - 15 min, acetonitrile 20% within 15 -30 min, acetonitrile 20% -30% within 30 -40 min. The flow rate was 1.0 mL · min- 1, the detection wavelength was 350 nm and the column temperature was 40℃. Results Different processing methods had certain effects on the content of chlorogenic acid and galuteolin inflos lonicerae. Conclusion It was suitable to use the processing of drying method and microwave in the production. And there was close relationship between the quality and appearance of color.
