| 摘 要： | 目的建立了一种简单的多西紫杉醇萃取方法和高效的反相高液相色谱法。方法血清中多西紫杉醇用乙醚-石油醚(9∶1)单步提取;色谱柱:Symmetry RP18(5μm,4.6mm×25mm,waters),流动相:甲醇-乙睛-水=45∶22∶33。检测波长230nm,室温,内标:地西泮。结果多西紫杉醇在0.25～12μg·mL-1范围内呈良好的线性关系(r=0.9993),平均回收率100.11%(n=9,RSD=2.50%)。结论此方法提取步骤少,方法简单快速、准确,适合临床多西紫杉醇的血药浓度监测。

| Abstract: | OBJECTIVE A simple extraction of docetaxel in serum and a method of HPLC were developed.METHODS docetaxel was extracted from serum by ethyl ether：petroleum ether（9∶1） and analyzed in a column of Symmetry RP18（5μm,4.6mm×25mm,waters） with a mobile buffer of methol-acetonitrile-water（45∶22∶23）.docetaxel was detected in 230nm with diazepam as internal standard in normal temperature.RESULTS The linear of docetaxel was excellent within the range of 0.5～12μg·mL-1（r=0.9993）.The average recoverys was 100.11%（n=9,RSD=2.50%）.CONCLUSION The method is simple,rapid,sensitive and applied to determine the therapeutic drug monitoring of docetaxel. |
