肼对大鼠心肌成纤维细胞增殖和凋亡影响的实验研究

摘    要:目的:不同浓度的肼处理体外培养的心肌成纤维细胞,观察其增殖和凋亡情况。方法:用MTT法检测对细胞增殖的影响;光学倒置相差显微镜和Hochest33258荧光染色观察细胞处理前后的形态学变化,AnnexinⅤ/PI双染流式细胞仪检测细胞凋亡率和坏死率。结果:MTT检测显示细胞增殖受到抑制;荧光染色后观察到明显的细胞凋亡的核浓缩、核聚集形态;流式细胞仪检测显示细胞凋亡率和坏死率与对照组比较差异均有显著性。结论:肼可抑制心肌成纤维细胞增殖,其作用随浓度增加而加强,低浓度(2mmol·L-1以下)的肼可导致心肌成纤维细胞凋亡,浓度大于2mmol·L-1则导致细胞坏死。

Abstract:Objective: To investigate the effect of hydrazine on cardiac fibroblast and to observe the apoptosis and proliferation of cardiac fibroblast induced by hydrazine. Methods: MTT assay was used for detecting the proliferation of cardiac fibroblast affected by hydrazine. The nucleus changes of apoptosis of cardiac fibroblast stained by Hochest33258 were observed through fluorescence microscope. The rate of apoptosis and necrosis of cardiac fibroblast stained by AnnexinⅤ-FITC/PI Apoptosis Detect KitⅠwas detected by flowcytometry. Results: Hydrazine inhibited the proliferation of cardiac fibroblast. The nucleus aggregation was detected in cardiac fibroblasts. The difference of the rate of apoptosis and necrosis was significant between the hydrazine-treated groups and the control. Conclusion: Hydrazine might inhibit the cardiac fibroblast proliferation and this role would strengthened with the increasing dose of hydrazine. Hydrazine might induce cardiac fibroblast apoptosis in a low dose, and the degree of apoptosis induced by 2 mmol·L -1 hydrazine was the most. The necrosis of cardiac fibroblast induced by over 2 mmol·L -1 hydrazine was much more.
