摘    要:目的:探讨急性髓系细胞白血病核基质蛋白(NMPs)的变化。方法:高盐抽提法制备白血病细胞NMPs。免疫动物制备多克隆抗体,以此作为免疫学探针对正常WBC和白血病细胞NMPs进行双向电泳(2DE)-免疫印迹(Western blot)检测。结果:在急性髓系细胞白血病细胞和正常WBC NMP8之间,有3个蛋白质表现出质和量上的差异。结论:2DE-Western blot方法能较好地分辨出正常WBC和白血病细胞NMPs的差异。

Abstract:Objective:To study the changes of nuclear matrix proteins(NMPs)in leukemia cells.Methods:NMPs were prepared by high salt extraction and immunized rabbit to raise polyclonal antibody which was used to detect the membrance transferred from 2 D gel of nuclear matrix in normal WBC and acute myeloid leukemia cells by Western blot.Results:There were 3 proteins spots appeared with difference between normal WBC and leukemia cells.Conclusion:2DE-Western blot is an available method to explore the difference between normal WBC and leukemia cells.
