摘 要: | 目的探讨肝核受体LXRs激动剂T0901317人表皮鳞状细胞癌SCL-1细胞增殖的影响及可能机制。方法 CCK-8法检测不同浓度T0901317(1.0、10.0、20.0μmol/L)作用SCL-1细胞24、48、72 h后细胞增殖率的变化。流式细胞仪检测不同浓度肝核受体激动剂对SCL-1细胞周期分布的影响。反转录-聚合酶链反应(RT-PCR)及免疫印迹试验(Western blot)检测肝核受体激动剂对细胞周期相关因子PCNA、p21、p27、CCND1表达水平的影响。结果 T0901317可明显抑制SCL-1细胞增殖,并呈现一定的时间和浓度依赖性;肝核受体激动剂作用于细胞株后,SCL-1细胞的G1期百分率上升,而S期、G2期百分率下降;且在一定浓度范围内导致细胞周期因子p21的m RNA及蛋白表达增加,而其他细胞周期相关因子表达未见明显变化。结论肝核受体LXRs激动剂可能通过促进细胞周期调节负性因子p21的表达,导致细胞周期静息甚至停滞于G1期,从而抑制细胞增殖。

Effects of LXRs agonists on the proliferation of SCL-1 cells |

Abstract: | Objective To detect the effects of LXRs agonists T0901317 on human cutaneous squamous cell carcinoma SCL-1. Methods 24,48 and 72 hours after T0901317(1.0, 10.0, 20.0 μmol/L) with different concentrations acting on SCL-1 cells, CCK-8kit assay was performed to detect cell proliferation. After having been treated with T0901317(1.0, 10.0, 20.0 μmol/L) for 24 hours, SCL-1 cells cycle progression was analyzed by flow cytometry. RT-PCR and Western blot were used to detect the expression of cell cycle related factors' genes and proteins. Results T0901317 inhibited the proliferation of SCL-1 cells in the concentration and time dependant manners. The proportion of SCL-1 cells in G1-phase increased, while that in G2- and S-phase decreased after having been treated with T0901317 (1.0, 10.0 ,20.0 μmol/L) for 24 hours. The result of PCR and Western blot showed that the expression of p21 protein was up-regulated, but the other cell cycle factors had no obviously changes. Conclusion The inhibition of SCL-1 cells proliferation by LXRs agonists is involved in G1-phase arrest, which might be related to the increased expression of p21 factor. |
