弓形虫病基因诊断的动物实验研究

摘    要:目的:建立敏感、特异、稳定的PCR法,用于弓形虫病诊断。方法:选择与合成引物并确立最佳扩增条件,用PCR法检测实验感染鼠血液中弓形虫DNA,并与ELISA法检测循环抗原(CAg)作比较。结果:该PCR检测弓形虫DNA法特异性强,对其他7种寄生虫DNA均不扩增;敏感性高,可检测到0.2pg DNA;检测急性感染弓形虫小鼠血液最早于感染后第2天出现用性,与ELISA法查CAg比较。PCK检测弓形虫DNA法阳性出现时间早、检出率高。结论:该PCR法可应用于弓形虫感染的早期诊断。

Abstract:Objective: To establish a sensitive, specific and stable PCR method to diagnose toxoplasmosis. Methods:After selecting and preparing primers and settling the best condition, the DNA of toxoplasma gondii in the blood of infected mice was detected by PCR and compared with the method of ELISA to detect circulating antigen(CAg) in the same mice. Results : The PCR method was very seasirive and specific,which could detect as little as 0.2 pg of DNA of T.gondii and there was no cross reaction with the DNAs of seven other parasites.The DNA could be detected as early as 2 days after the infection, which was much earlier than by ELISA. Conclusion:The PCR method can be applied in the early diagnosis of toxoplas-mosis.
