鼻咽癌原发灶与颈部淋巴结转移癌组织中nm23-H1、MMP-9蛋白表达及其意义

摘    要:目的:研究鼻咽癌原发灶和颈部淋巴结转移癌组织中nm23-H1(non-metastatic clone No.23)、MMP-9(matrix metalloproteinase9)基因蛋白的表达及其意义。方法:采用免疫组化二步法检测22例鼻咽癌患者自身原发灶和颈部淋巴结转移癌组织中nm23-H1、MMP-9蛋白的表达。结果:淋巴结转移癌组织的nm23-H1阳性表达率(17/22,77.3%)低于原发灶组织中的表达水平(18/22,81.8%),但未见显著性(P>0.05);MMP-9在淋巴结转移癌组织中,癌细胞阳性表达率(19/22,86.4%)高于原发灶组织中的表达水平(13/22,59.1%),差异有显著性(P<0.05);原发灶与转移灶中nm23-H1呈正相关,转移癌组织中nm23-H1和MMP-9呈负相关,均有统计学意义(P<0.05)。结论:在鼻咽癌组织中,nm23-H1蛋白的表达下调可能是导致癌细胞侵袭转移的一个重要因素。MMP-9在鼻咽癌组织其表达强度可能与鼻咽癌病期进展和颈部淋巴结转移有关。

Abstract:Objective:To study the expressions and their clinical significance of nm23-H1 and MMP-9 in primary nasopharyngeal carcinoma(NPC)and cervical lymph node metastasis carcinoma.Methods:Envision immunohistochemical staining was performed in 22 cases self-control of NPC and cervical lymph node metastasis carcinoma.Results:The positive expression rate of nm23-H1 in cervical lymph node metastasis carcinoma was lower than that in NPC(77.3%,81.8%),but without significance(P<0.05).A negative correlation between the two groups.On the other hand,the MMP-9 positive expression in lymph node group was stronger than that of NPC(86.4%,59.1%),and a significant difference was found between them(P>0.05).A positive correlation was found between the nm23-H1 in NPC and metastasis carcinoma.While in lymph node group,a negative correlation was found between nm23-H1 and MMP-9.Conclusion:The down regulation of nm23-H1 might play a role in the NPC invasion process.MMP-9 may be one of the biological criterions of NPC malignancy,as its expression might be correlated with NPC advancement and cervical lymph node metastasis.
