不同来源TIMP-1基因表达对人乳腺癌MDA-MB-231细胞生物学行为影响的研究

摘    要:目的研究不同来源金属蛋白酶1组织抑制剂(TIMP-1)对人乳腺癌MDA-MB-231细胞侵袭与转移特性的影响。方法分别构建内源性和外源性TIMP-1表达乳腺癌细胞模型,检测MDA-MB-231细胞、MDA-MB-231(TIMP-1)细胞和MDA-MB-231(AdTIMP-1)细胞在体外增殖速度、克隆形成、重组基底膜侵袭抑制试验、细胞移动试验和SCID免疫缺陷小鼠移植瘤形成试验进行体内外研究。结果通过细胞体外增殖速度检测显示,内源性TIMP-1的表达有可能促进肿瘤细胞的生长;外源性TIMP-1的表达有可能抑制肿瘤细胞的生长。无论是内源性还是外源性的TIMP-1表达均不影响肿瘤克隆形成能力和肿瘤形成能力。内源性TIMP-1的表达有可能促使肿瘤细胞的移动能力增强;外源性TIMP-1表达不影响肿瘤细胞的移动能力。内、外源性TIMP-1对MDA-MB-231的侵袭能力均有抑制作用,但外源性TIMP-1的抑制作用更大。通过体内致瘤实验显示,内源性TIMP-1的表达可显著促进肿瘤生长;外源性TIMP-1的表达可显著抑制肿瘤生长。结论不同来源的TIMP-1对MDA-MB-231细胞的生物学行为有不同影响。

Abstract:Objective To study the effect of different TIMP-1 resources for invasive and metastatic characteristics of MDA-MB-231 cell. Methods The models of breast cancer cells of the endogenous and exogenous TIMP-1 expression were established separately. Detecting MDA-MB-231 cell, MDA-MB-231 (TIMP-1) cell, and MDA-MB-231 (AdTIMP-1) cell in proliferative rate in vitro, clone formation, invasive and inhibitive test of remodeling basal membrane, cell movement test, and transplanted tumorigenesis test of SCID immunodeficient mice in vivo or in vitro. Results Endogenous TIMP-1 expression might promote the growth of tumor cells; Exogenous TIMP-1 expression might inhibit the growth of tumor cells. Neither endogenous nor exogenous TIMP-1 expression could influence the ability of tumor cloning formation and tumor formation. Endogenous TIMP-1 expression might promote the increasing ability of movement in tumor cells but exogenous TIMP-1 expression could not influence the ability of movement in tumor cells. Endogenous and exogenous TIMP-1 had inhibitive activities for the invasive ability of MDA-MB-231 cells,however,exogenous TIMP-1 had much more inhibitive activity than endogenous one. Endogenous TIMP-1 expression could promote the growth and metastasis of tumor obviously. Exogenous TIMP-1 expression could inhibit the growth of tumor obviously. Conclusions TIMP-1 from different resources have different effects for biological conducts of MDA-MB-231 cells.
