Axin基因在脑肿瘤干细胞和神经干细胞中的表达及其多态性研究

摘    要:目的观察Axin基因在脑肿瘤干细胞和神经干细胞中的表达及其多态性,为临床治疗脑肿瘤疾病提供实验依据。方法取大鼠神经干细胞及胶质瘤肿瘤干细胞,采用RT-PCR、基因测序等方法研究Axin基因在脑肿瘤干细胞和神经干细胞中的表达,对反应产物进行测序,观察两组之间的序列差异,并与Genebank Axin基因序列进行比较。结果相差显微镜下观察,呈悬浮状态的脑肿瘤干细胞聚集在一起的小球,随着培养时间的延长,小球数量增多,球体不断增大但并不均一,长出的新球体大小均一,数量多。分离原代的神经干细胞形成细胞球。脑肿瘤干细胞中Axin序列的表达与神经干细胞及Genebank中Axin比较差异性很大,其相似性为75.6%。结论Axin基因在神经干细胞中正常表达,而在脑肿瘤干细胞中表达过低,说明Axin在脑肿瘤细胞增殖中起着重要的抑制作用。

Abstract:Objective To explore the expression and polymorphism of Axin in brain tumor stem cells and neural stem cells. To provide an evidence for clinical diseases. Methods Choose the neural stem cells and aglioma stem cells, investigate expression Axin in brain tumor stem cells and neural stem ceils by RT-PCR and DNA Sequencing, contrasted to Genebank Axin. Results The cells that were isolated from brain tumors and the neural stem cells form neurospheres. The expression comparability of Axin is 75.6% in brain tumor stem cells and neural stem cells. Conclusion Axin gene could express normally in neural stem ceils and very low in aglioma stem cells.
