硫唑嘌呤致小鼠急性肝组织损伤模型的建立

摘    要:[目的]探讨硫唑嘌呤(AZA)诱导小鼠制作急性肝组织损伤模型的最佳条件.[方法]分别灌胃给予各组小鼠50,100,150,200mg/LAZA,给药后24h测定血清中AST,ALT值;分别灌胃给予各组小鼠150mg/LAZA,给药后4,8,12,16,20,24,28,32h测定血清中AST,ALT值.[结果]随着给予AZA剂量的增加,小鼠血清中AST,ALT值升高;随着给药时间的延长,小鼠血清中AST,ALT值在12h时最高,之后逐渐下降.[结论]利用AZA制作急性肝组织损伤小鼠模型时,AZA的最佳给药剂量为150mg/kg,最佳作用时间为给药后12h.

Abstract:OBJECTIVE To investigate the optimal condition to induce acute hepatic injury model in mice by azathioprine(AZA).METHODS Mice in per groups were given 50,100,150,200 mg/L dose of AZA for 24 hours,then the contents of AST,ALT in serum were measured.Mice in per groups were given 150 mg/kg dose of AZA,after 4,8,12,16,20,24,28,32 hours,the contents of AST,ALT in serum were measured too.RESULTS The levels of AST,ALT in serum in AZA-treated mice were dose-dependently increased,and those reached maximum peak at 12 hours,then decreased after then.CONCLUSION The optimal condition to make the acute hepatic injury model induced by AZA in mice are administrated 150 mg/kg AZA and treated for 12 hours.
