大肠杆菌O—157产生的VT2对中性粒细胞自发性凋亡的抑制作用

摘    要:目的 研究毒源性大肠杆菌(VETC)O-157产生的外毒素-VT2对中性粒细胞自发性凋亡的影响,探索VET感染后并发的溶血性尿毒症(HUS)及相关疾病的发病机理。方法 运用形态学观察、流式细胞仪(FACS)技术和DNA分离技术进行研究。结果 在无VT2存在的情况下,经过24h体外培养,65%左右的中性粒细胞发生了凋亡,而浓度为10^-6M的VT2使其凋亡发生率降至20%左右。结论 VT2延长了中性粒细胞的功能性生命期限,使其胞浆内细胞毒性物质有更多的机会损伤肾小球血管内皮细胞,促进微血栓的形成,因而中性粒细胞可能参与了HUS的形成过程。

Abstract:Aim To evaluate the pathological role of verotoxin 2(VT2)in the hemolytic uremic syndrome (HUS )and VETC -associated diseases,we investigated the effects of VT2on the neutrophil apoptosis.Methods The observation of morphological ch anges of neutrophils,FACScan flow cytometer and technique of DNA separation were used in this study.Results VT2at the concentration of 10 -6 M significantly inhibited spon-taneous apoptosis of neutrophils(from 65%in negative control to 20%in VT2).Conclusion VT2prolonged the functional life -s pan of neu-trophils so that the cytotoxic conte nts of neutrophils may have more chances to destroy glomerular micro vascular endothelial cells,suggesting that the neutrophils affected by VT2might be involved in HUS and VETC -associated diseases.
