柴胡疏肝散含药血清对衣霉素诱导内质网应激神经元轴突生长的影响

摘    要:目的:探讨柴胡疏肝散含药血清对衣霉素(TM)诱导的NG108-15细胞轴突生长的影响。方法:实验采用衣霉素对NG108-15神经细胞进行孵育,诱导其形成内质网应激(ERS)细胞模型,分别用正常兔血清、安理申含药兔血清、高、中、低剂量柴胡疏肝散含药血清进行干预,观察各组细胞轴突生长情况。结果:加入柴胡疏肝散含药血清高、中、低剂量孵育干预后,细胞的突起阳性率和平均突起长度明显优于模型组(P0.01);高剂量组柴胡疏肝散含药血清与低剂量组相比较(P0.05),与西药安理申组含药血清两者之间无明显差异(P0.05)。结论:柴胡疏肝散含药血清可促进衣霉素诱导NG108-15细胞内质网应激模型神经元突起的生长。

Effect of Chaihushugansan Medicated Serum on Neuron Axons Growth of Endoplasmic Reticulum Stress Cell Model Induced by Tunicamycin

Abstract:Objective To observe the effect of Chaihushugansan(CHSGS) on neuron axons growth ofNG108-15 endoplasmic reticulum stress cell model induced by tunicamycin (TM). Methods The endoplasmic reticulum stress(ERS) NG108-15 cell models were induced by tunicamycin(TM), and then were divided into five groups which were respectively incubated with normal rabbit serum, rabbit serum containing Aricept, rabbit serum containing high, middle and low dose CHSGS. The neuron axons growth statuses of all groups were observed. Results After incubating with different medicated serum, the neurite positive rates and average neurite lengthes of different dose of CHSGS were significantly better than that of the ERS model group (P 〈 0.01); and there was an obvious difference between the high dose CHSGS group and low dose CHSGS group (P 〈 0.05), but no obvious difference between the high dose CHSGS group and the aricept group (P 〉 0.05). Conclusions The medicated serum of CHSGS could increase the number and the length of the axons of ttmicamycin induced NG108-15 ERS cell model.
