痛泻要方及防风对肠上皮Caco-2细胞屏障功能的影响

摘    要:目的:研究痛泻要方及防风对肠上皮Caco-2细胞屏障功能的影响。方法:应用蛋白酶激活受体2(PAR-2)激动剂(胰蛋白酶)诱导Caco-2细胞建立肠上皮细胞屏障功能障碍模型,以跨膜电阻(TEER)和荧光黄透过率为指标,评价其屏障通透性。分别给予含痛泻要方原方、痛泻要方无防风方、防风的血清研究药物对Caco-2细胞屏障通透性的影响。结果:痛泻要方和防风均显著升高Caco-2细胞屏障的TEER值,降低细胞屏障荧光黄透过率(P0.05),痛泻要方无防风方对上述指标无显著影响(P0.05)。结论:痛泻要方及防风对肠上皮Caco-2细胞屏障起保护作用。

Abstract:Objective To study the effect of Radix Saposhnikoviae and Tongxie Yaofang on the barrier functions of human intestinal Caco-2 cell. Methods The model of the intestinal epithelial barrier was established by cultivating Caco-2 cell in the Transwells and was induced by trypsin which was a protease activated receptor2(PAR-2) agonist.The effects of Tongxie Yaofang(TXYF), TXYF exluding Radix Saposhnikoviae (RS),RS on the permeability barrier model were evaluated by measuring transepithelial electrical esistance(TEER)and transepithelial transmittance offluorescein. Results TXYF and RS significantly increased TEER values of permeability barrier model(P 〈 0.05), there was no significant effect of TXYF exluding RS(P 〉 0.05). TXYF and RS significantly decreased the transepithelial transmittance offluorescein(P 〈 0.05). Conclusion TXYF and RS play an important action on protecting the intestinal epithelial barrier.
