百合地黄汤对失眠模型大鼠行为学及不同脑区单胺类神经递质的影响

摘    要:目的:研究百合地黄汤对失眠模型大鼠行为学及大鼠皮层、脑干和海马组织中多巴胺(DA)和5-羟色胺(5-HT)的影响,探讨百合地黄汤治疗失眠的作用机制。方法:采用腹腔注射对氯苯丙氨酸(PCPA,0.4 g·kg-1)复制失眠模型大鼠,将造模成功的大鼠随机分为模型组、地西泮组(0.92 mg·kg-1)、百合地黄汤高剂量组(28 g·kg-1)和百合地黄汤低剂量组(7 g·kg-1),另设空白组,连续给药7天,采用旷场实验测定行为学评分,翻正反射实验评价百合地黄汤对大鼠睡眠的影响,高效液相色谱电化学检测法(HPLC-ECD)检测大鼠皮层、脑干和海马中DA和5-HT的含量。结果:百合地黄汤可以降低失眠模型大鼠的中央格停留时间、修饰次数和粪便颗粒数(P < 0.05),缩短睡眠潜伏期并延长睡眠持续时间(P < 0.05),大脑皮层、脑干和海马中5-HT含量明显升高(P < 0.05),大鼠皮层中DA含量显著降低(P < 0.05);与模型组相比,P < 0.05。结论:百合地黄汤可改善失眠模型大鼠行为学和睡眠,对DA和5-HT的调节作用可能是其治疗失眠的作用机制。

Abstract:Objective: To study the effects of Baihe Dihuang Decoction on the ethology of insomnia model rats and dopamine (DA) and serotonin (5-HT) in the cortex, brain stem and hippocampus of rats, so as to study the mechanism of BaiheDihuang Decoction in the treatment of insomnia. Methods: Rats were given parachlorophenylalanine (PCPA, 0.4 g·kg-1)to replicate insomnia model, and were randomly divided into model group, diazepam group (0.92 mg · kg- 1), Baihe Dihuang Decoction high dose group (28 g·kg-1) and Baihe Dihuang Decoction low dose group (7 g·kg-1), and blank group.All groups were administrated continuously for 7 days, The behavior score was measured by open field test, and thesleeping of rats was evaluated by reversal reflex experiment. The contents of DA and 5-HT in the cortex, brain stem andhippocampus of rats were measured by high performance liquid chromatography with electrochemical detection method(HPLC -ECD). Results: Baihe Dihuang Decoction can reduce the time in center grid, the number of fecal particles andgrooming (P < 0.05), shorten the sleep latency and prolong the duration of sleeping quality (P < 0.05), and the 5-HTcontent in the cortex, brain stem and hippocampus of insomnia model rats increased significantly (P < 0.05). The contentof DA in rat cortex decreased significantly (P < 0.05), compared with the model group, P < 0.05. Conclusion: BaiheDihuang Decoction can improve the ethology and sleeping quality of insomnia rats, and its regulating effect on DA and 5-HT may be the mechanism of insomnia treatment.
