腹水miR-4455和miR-29a在结核和肿瘤中的表达差异研究

目的：探讨微小RNA（microRNA,miR）在结核性腹水和肿瘤性腹水中的表达差异。方法：收集腹水标本提取RNA,Taq-Man定量PCR方法检测miR-4455、miR-29a的表达。结果：肿瘤性腹水中miR-4455的表达较结核性腹水增高（P〈0.05）、而miR-29a的表达较结核性腹水降低（P〈0.05）;ROC曲线分析显示miR-4455、miR-29a及二者联合检测对结核性腹水和肿瘤性腹水鉴别诊断的AUC值分别为0.882（95%CI：0.823-0.931）、0.842（95%CI：0.774-0.903）和0.928（95%CI：0.872-0.956）。结论：腹水miR-4455、miR-29a的表达在结核性腹水和肿瘤性腹水中存在明显差异,可能成为二者鉴别诊断的标记物。

Differential expression of ascites miR-4455 and miR-29a clistinguishing tuberculosis and cancer

Objective： To explore the differentail expression of microRNAs in tuberculous and cancerous ascites. Methods： Ascites samples were collected for total RNA extraction. The TaqMan quantatitive PCR method was used to assess the expression of miR-29 a and miR-4455. Results： Ascites miR-4455 was higher while miR-29 a was lower in patients with cancer than in patients with tuberculous peritonitis（P〈 0.05）.Receiver operating characteristic（ROC） curve analysis showed that the value of AUC,which distinguishing tuberculous peritonitis from malignant ascites,was from 0.882 and 0.842 increased to 0.928 when the two miRs.Conclusion： Altered expression of miR-4455 and miR-29 a in ascites might be a novel biomark for the differential diagnosis of tuberculous ascites from cancer ous ascites.
