硫酸软骨素酶ABC对大鼠脊髓损伤后硫酸软骨素蛋白多糖与胶质纤维酸性蛋白表达的影响

摘    要:目的探讨硫酸软骨素酶ABC对严重脊髓损伤(SCI)大鼠硫酸软骨素蛋白多糖(CSPGs)及胶质纤维酸性蛋白(GFAP)表达的影响。方法制作大鼠脊髓横断伤模型,60只大鼠随机分为对照组10只、损伤组25只和治疗组25只。治疗组给予硫酸软骨素酶ABC髓鞘浸润治疗。用免疫荧光组织化学观察脊髓CSPGs的表达;用免疫荧光组织化学及Western-blot方法观察脊髓GFAP的表达。结果 SCI后星形胶质细胞明显活化增殖,CSPGs和GFAP的表达明显增加(P<0.05);治疗组脊髓CSPGs和GFAP的表达均较损伤组减少,差别均有统计学意义(P<0.05)。结论硫酸软骨素酶ABC能够裂解CSPGs,改善SCI区的微环境,改善GFAP蛋白的表达,是修复SCI,促进神经再生的机制之一。

Abstract:Objective To investigate the effects of chondroitinase ABC on the expression of chondroitin sulfate proteoglycans ( CSPGs) and glial fibrillary acid protein ( GFAP) after spinal cord injury ( SCI) in rats. Methods The models of rat spinal cord transsection were established. Sixty rats were randomly divided into control group( n = 10) ,SCI group( n = 25) and treatment group( n = 25) . The rats in treatment group were treated with chondroitinase ABC infiltrating through medullary sheath. The expression of CSPGs in spinal cord was observed by immunofluorescent histochemical staining,the expression of GFAP in spinal cord was observed by immunofluorescent histochemical staining and Western-blot. Results After SCI,the horizontal cells were activated and proliferated,and the expression of CSPGs and GFAP was significantly increased( P < 0. 05) . The expression of CSPGs and GFAP in treatment group was significantly less than those in SCI group( P < 0. 05) . Conclusion Chondroitinase ABC can split CSPGs. It can improve the microenvironment of the injured,and enhance the expression of GFAP,which is one of the mechanism of repairing the SCI.
