反式BPDE诱发的转化细胞与成瘤细胞FHIT基因变化研究

摘    要:目的:探讨苯并(a)芘[B(a)P]代谢产物反式二氢二醇环氧苯并芘[anti-7,8,-dihydrodiol-9,10-epoxidebenzo(a)pyrene,BPDE]引起的细胞FHIT基因突变情况。方法:PCR、RT-PCR、多聚酶链聚合反应-单链构象多态性分析(PCR-SSCP)。结果:PCR-SSCP结果提示反式BPDE诱发的恶性转化细胞及其裸鼠成瘤细胞FHIT基因的Exon1-9出现了异常的小片断。结论:反式BPDE可作用于细胞染色体3p14.2的脆性部位FRA3B,引起FHIT基因的重排或缺失,导致FHIT基因功能失活。推测FHIT基因可能是反式BPDE致癌作用的靶分子之一。
