摘    要:目的研究二烯丙基二硫(DADS)诱导胃癌BGC823细胞凋亡及其机制。方法实验分DADS处理组和DADS未处理组.进行体外细胞培养,通过MTr法检测DADS对胃癌BGC823细胞生长的影响、通过流式细胞术检测细胞周期的分布、通过免疫组化检测smac和survivin蛋白的表达。结果MTT实验显示DADS作用于BGC823细胞后,细胞生长抑制率呈时间、浓度依赖性增高,流式细胞术显示DADS明显将细胞周期阻滞于G2/M期,且呈浓度、时间依赖性。免疫细胞化学显示处理组较未处理组snlae表达明显增加,而survivin蛋白表达明显降低(P〈0.05)。结论DADS可体外抑制人胃癌BGC823细胞增殖,使该细胞阻滞于G2/M期,其机制可能与上调smac及下调survivin的表达有关。

Abstract:Objective To investigate the apoptosis in human gastric cancer BGC823 cells induced by DADS,and study the mechanism.Methods The experiment was divided into two groups: treated group with DADS and untreated group.BGC823 cells proliferation was measured by MTT assay.Cell cycle distribution was analyzed by flow cytometry(FCM).The expression of survivin,smac was detected by immunohistochemistry.Results MTT showed that DADS significantly inhibited BGC823 cells proliferation with a dose and time-dependent manner,Flow cytometry revealed that DADS could markedly arrest cell cycle in G2/M phase with a dose and time-dependent manner.The expression of survivin and smac were observed in the two groups of immunohistochemistry,survivin was weakened markedly in the treated group,but smac was enhanced markedly in the treated group(P < 0.05).Conclusion DADS had significant inhibitory effect on proliferation of human gastric cancer BGC823 cells in vitro and arrested cell cycle in G2/M,and its molecular mechanism is related to downregulating expression of survivin and upregulating expression of smac.
