黄芪提取物联合表皮生长因子治疗大鼠皮肤烫伤的实验研究

摘    要:目的:探讨黄芪提取物联合表皮生长因子对皮肤烫伤愈合的疗效。方法:Wistar大鼠80只,每只背部造成2处直径2.8cm深Ⅱ度烫伤创面。将大鼠随机分为黄芪提取物联合表皮生长因子治疗组(联合组)、表皮生长因子治疗组(EGF组)、黄芪提取物治疗组(黄芪组)和生理盐水对照组(对照组),每组20只,观察伤后愈合情况。烫伤后第2、5、7、10、14、18、21天计算创面愈合率;第7、14、18天取大鼠创面皮肤采用羟脯氨酸测试盒测定烫伤创面羟脯氨酸含量;第7、14、21天取大鼠创面皮肤采用Ⅷ因子免疫组织化学染色计算微血管密度;第14、21天取大鼠创面皮肤做病理学分析。结果:联合组愈合时间显著小于其它组(P〈0.05,P〈0.01),EGF组和黄芪组愈合时间显著小于对照组(P〈0.05);联合组于烫伤后10、14、18、21d和7、14d创面愈合率、创面微血管密度明显大于其它组(P〈0.05,P〈0.01),EGF组和黄芪组创面愈合率以及创面微血管密度显著大于对照组(P〈0.05);联合组创面组织羟脯氨酸水平在伤后第7、14天均显著高于其他组(P〈0.05,P〈0.01),EGF组和黄芪组创面组织羟脯氨酸水平显著高于对照组(P〈0.05)。结论:黄芪提取物联合表皮生长因子能显著促进皮肤烫伤的愈合,缩短愈合时间。

Study of extract of astragalus united epidermal growth factor for the treatment of rat skin burns

Abstract:AIM:To investigate the effects of extract of astragalus united epidermal growth factor(EGF)on skin burns healing efficacy.METHODS:80 Wistar rats with each back,2.8 cm in diameter caused by two deep Ⅱ degree burn wound.Rats were randomly divided into extract of astragalus united epidermal growth factor treatment group(United group),epidermal growth factor treatment group(EGF group),extract of astragalus treatment group(RA group),and isotonic saline control group(control group),each 20 to observe the healing after injury.The wound healing rate after 2,5,7,10,14,18,21 days was measured and calculated.In scalded 7,14,18 days,the content of wound hydroxyproline was measured by hydroxyproline test kit.In scalded 7,14,21 days,the microvessel density wound was calculationed by use of Ⅷ immunohistochemical staining.The 14,21 days,the rat skin wound was taken to do pathology analysis.RESULTS:The healing time of the United group were significantly lower than those in the other groups(P0.01,P0.05).Compared with control group,the healing times of EGF group and RA group were lower(P0.05).The wound healing rate,the wound microvessel density were higher in United group than those in the other groups(P0.01,P0.05)on scalded 10,14,18,21 days and in 7,14 days.Compared with control group,the wound healing and wound microvessel density in EGF group and RA group were higher(P0.05).The content of wound hydroxyproline in United group were significantly higher than those in the other groups(P0.01,P0.05)on scalded 7,14 days.Compared with control group,the content of wound hydroxyproline in EGF group and RA group were higher(P0.05).CONCLUSION:The extract of astragalus united epidermal growth factor can significantly promote skin wound healing and shorten the healing time.
