摘    要:考察不同流动相pH、色谱柱和柱温对奥赛博司RP—HPLC色谱行为的影响。方法:色谱柱:Waters SunFire和Kromasil 100—5RPC18(500mm×4.6mm,5μm);流速:1ml·min^-1;检测波长:220nm;进样量:20μ1;柱温:(25,28,30,35℃);流动相:磷酸盐缓冲溶液(0.1mol·L^-1。KH2PO4+0.02mol·L^-1H3PO4)乙腈=8:2(磷酸盐缓冲溶液用磷酸二氢钾饱和溶液调pH分别为:2.83,2.84,2.95,2.99,3.02,3.05,3.07,3.14,4.0,4.50,5.0,5.50,6.05)。结果:两种色谱柱、不同柱温对奥赛博司的色谱行为没有太大影响;但当流动相pH不同时对其有一定影响。结论:本方法可为奥赛博司质量标准的系统研究提供依据。

Abstract:To study the effect of various pH value of mobile phase, different columns and column temperature on the chromatographic behavior of OSAIBOS. Method: The chromatographic separation was performed on RPC18 (500 mm×4.6ram ,5μm) of Waters SunFire and Kromasil 100-5 with the column temperature at 25, 28, 30, 35℃ respectively and detection wavelength at 220nm, the flow rate was set at 1 ml·min^-1 and 20μ1 of sample was injected for analysis; the mobile phase consisted of a phosphate buffer solution (0.1 mol·L^-1 potassium dihydrogen phosphate +0.02 mol·L^-1 Phosphoric acid adjusted pH with potassium dihydrogen phosphate saturated solution to 2.83,2.84,2.95,2.99,3.02,3.05,3.07,3.14,4.0,4.50,5.0,5.50,6.05 respectively) : acetonitrile = 8: 2. Result: These two chromatographic columns and different column temperature have no significant influence on the chromatographic behavior of OSAIBOS, but the pH value of mobile phase does. Conclusion : This method can provide bases for the systematic study of quality criteria of OSAIBOS.
