Abstract:Objective To study the neuroprotective effect of the ischemic postconditioning method on spinal cord isehemia reperfusion injury and the possible mechanism related to phosphatidylinnositol 3 kinase-Akt (PI3K/Akt). Methods Forty two Japanese white rabbits weighing between 2.0-2.5 kg were randomly divided into seven groups: isehemic group(group I), isehemic postconditioning group(group PB), PB+DMSO group ( group D ) , PB+ LY294002 5 μg group ( group PY5 ) , PB+ LY294002 10μg group ( group PY10 ) , PB+ LY294002 20μg group ( group PY20 ) , LY294002 10 μg group ( group LY ) . Spinal cord ischemia was induced in rabbits by occlusion of the infrarenal aorta with a balloon catheter for 25min. During the first 10 min of reperfusion, blood flow was partially restored and the mean blood pressure of the distal aorta was controlled between 45 and 55 mmHg by adjusting the volume of the balloon in the postconditioning group. Functional evaluation with the Tarlov score was assessed 24h after spinal cord ischemia. Expressions of phospho-Akt and Bcl-2, Bax in the spinal cord were evaluated by Western blot. Results Compared with the ischcmia, the Tarlov scores was markedly increased 24h after spinal cord iscbemia with postconditioning method. The postconditioning group enhanced the expressions of phospho-Akt and Bcl-2 in spinal cord, but decreased the expressions of Bax (P〈 0.05 ). The neuroproteetive effect and the increases in phospho-Akt were abolished by administration of PI3K/Akt inhibitor LY294002. Conclusions The neuroprotective effect of the ischemic postconditioning on spinal cord ischemia reperfusion injury might be related to ohosohatidvlinnositol 3 kinase-Akt (PI3K/Akt) signal nathwav.
