猪皮活性小肽对L929细胞增殖的研究

目的 通过大孔吸附树脂DA201-C对猪皮活性小肽溶液的精制分离,研究猪皮活性小肽不同洗脱部位对小鼠成纤维细胞(简称L929细胞)增殖作用的影响.方法 采用仿生酶解法得到猪皮活性小肽溶液;采用超滤法得到分子量<3KDa的猪皮活性小肽超滤液;以大孔吸附树脂DA201-C对猪皮活性小肽超滤液进行分离纯化;以体外培养L929细胞为研究对象,探究猪皮活性小肽不同洗脱部位对L929细胞增殖的作用,用MTT比色法检测细胞增殖活性.结果 猪皮活性小肽不同洗脱部位对L929细胞均具有极显著的增殖作用,其中水洗脱组的活性最强.结论 猪皮活性小肽对L929细胞具有显著的增殖作用,可作为活性原料在皮肤创伤等领域广泛应用.

Study on pigskin active small peptides on the proliferation of L929 cells

Objective Refine and separate active small peptides solution of pigskin with macroporous adsorption resin DA201-C, different fractions of small peptide on pigskin effect activity of mouse fibroblast cells (hereinafter referred to as L929 cells proliferation). Methods Get the pigskin active small peptides solution by using the bionic enzymatic method; get the pigskin active small peptide with molecular weight of <3KDa ultrafiltration by ultrafiltration; refine and separate active small peptides solution of pigskin with macroporous adsorption resin DA201-C; regarded in vitro cultured L929 cells as the research object to explore different fractions of active small peptides elution of pigskin on the pro-liferation of L929 cells by MTT colorimetry.Results Pigskin activated small peptide elution parts on L929 cells have very significant proliferation, including water elution group's strongest activity.Conclusion Pigskin active small peptide could significantly promote the proliferation of L929 cells, as active ingredients in the treatment of skin wounds, and other fields have broad application prospects.
