胃癌组织中MUC1表达的意义

目的:检测MUC1在胃癌及胃良性病变组织中的表达,探讨其在胃癌诊断和免疫治疗中的意义.方法:采用免疫组化方法检测58例胃癌及26例胃良性病变组织、10例胃正常组织中MUC1的表达.结果:胃癌组织中MUC1阳性表达46例,为深棕色或棕黄色颗粒,主要位于细胞核上;良性病变组织中4例可见弱阳性反应信号;正常胃组织中未见MUC1表达.MUC1在胃癌组织中的阳性表达率与胃良性病变组织及正常组织之间存在显著性差异(P＜0.05).MUC1基因表达与组织学分化及部位无关(P＞0.05);与组织浸润转移及与临床分期有关(P＜0.05).结论:MUC1在胃癌组织中的表达及分布特点可作为胃癌的鉴别诊断指标,有可能作为胃癌免疫治疗的靶分子.

Expression significance of MUC1 in gastric carcinoma tissue

AIM: To investigate the differences of MUC1 expression in gastric carcinoma, benign gastric diseases and normal tissue and to further explore its clinical significance. METHODS: The expression levels of MUC1 were examined by a detailed immunohistochemical analysis with MUC1 monoclonal antibody in 58 samples of gastric carcinoma, 26 samples of benign diseases tissues and 10 normal tissues. RESULTS: Immunohistochemical analysis showed that positive expression of MUC1 was detected in 46 cases of gastric carcinoma, which aberrantly localized in the cancer cell nucleus, and weak positive expression in 4 cases in different benign diseases tissues. There was no expression of MUC1 in normal gastric tissue. The expression level of MUC1 in gastric carcinoma was much higher compared with that in benign and normal tissue and there was a significant difference between the expressional levels in gastric carcinoma and those in benign and normal tissue ( P <0.05). The positive expression levels of MUC1 in gastric carcinoma had no statistically significant correlation to the histological grade and location ( P >0.05) but were significantly correlated to the clinical stage and lymph node metastasis ( P < 0.05). CONCLUSION: The results suggest that the over expression and aberrantly localization of MUC1 in gastric carcinoma be used as a differentiating diagnostic marker of gastric carcinoma, and as the target of immunotherapy of gastric carcinoma.
