| 摘 要： | 作者应用纯化的前列腺酸性磷酸酶(PAP)免疫BALB/c小鼠,制备了4株分泌PAP-McAb的杂交瘤细胞株。应用PAP-McAb建立了免疫化学法测定血清中的PAP。318例正常男性,血清中PAP为0～1.6ng/ml。9例病理确诊的前列腺癌。有7例阳性,阳性率为77.8％,而同时应用化学法测定血清中的酸性磷酸酶(ACP),阳性率只有44.4％。 |

| Abstract: | Prostatic acid phosphatase (PAP) was purificated according to Skinnigsr-und's method which was modified. BALB/c mice were immunized with the mixture of PAP and complete Freund's adjuvant and spleen cells from a boosted mouse were fused with Sp2/0 mouse myeloma line. Four hybridoma cell lines secreting specific monoclonal antibodies against PAP were established. Immunochemistry assay has been developed for detection of serum PAP. It was shown that normal value of serum PAP (318 men) was 0-1.6ng/ml. The positive rates in prostatic cancer was 77.8%(7/9). On the contrary, the positive rate of chemical determination of serum acid phosphatase was only 44%. It suggests that McAb immunochemistry assay is specific sensitive and it can be used for determination of serum PAP. |
