氨基葡糖及其盐酸盐诱导白血病细胞K562向巨噬细胞样分化

摘    要:目的 研究氨基葡萄糖及其盐酸盐对白血病细胞K5 6 2的诱导分化效果。方法 利用hexamethylenebisac etamide (HMBA)、氨基葡萄糖及其盐酸盐 ,分别用不同浓度对白血病K5 6 2细胞进行诱导分化实验 ,通过联苯胺、萘酚AS D氯醋酸酯酶及吉姆萨 瑞氏染色、电镜观察和流式细胞仪证实其分化方向。结果 氨基葡萄糖及其盐酸盐对K5 6 2细胞在 0 5mmol·L-1浓度作用 2d后有良好的诱导分化效果 ,诱导K5 6 2向巨噬细胞分化并能使K5 6 2细胞周期阻滞于G0 /G1期 ,与HMBA相比 ,氨基葡萄糖和盐酸盐在作用浓度和作用时间上都有明显优势。结论 氨基葡萄糖及其盐酸盐都可能成为新的高效、低毒的治疗髓系白血病的候选药物

Abstract:AIM To study the differentiation effect of leukemia cell line K562 induced by glucosamine and glucosamine hydrochloride. METHODS K562 cell were induced differentiation by HMBA and glucosamine and glucosamine hydrochloride in different concentrations. The differentiation model were evaluated by the staining of benzidine, naphthyl AS D chloroacetate esterase, Wright Giemsa and electron photomicrograph, fluorescence activated cell analyzer (FACS). RESULTS Glucosamine and glucosamine hydrochloride induced K562 cell line into differentiation in the concentration of 0 5 mmol·L -1 after treated 2 days. Compared with HMBA, glucosamine and glucosamine hydrochloride have their advantages in the treatment concentration and time. CONCLUSION Glucosamine and glucosamine hydrochloride may became the high potency and low toxicity candidate drug in treatment of leukemia.
