壬基苯酚对豚鼠心室肌细胞L-型钙电流的影响

摘    要: 目的观察环境激素壬基苯酚(NP)对豚鼠心室肌细胞L-型钙电流(ICa-L)的影响。方法应用酶机械法分离豚鼠单个心室肌细胞;应用膜片钳全细胞方法观察NP对单个心室肌细胞ICa-L的影响。结果NP可抑制不同膜电位下的ICa-L,NP(10-6mol·L-1、10-5mol·L-1)使ICa-L峰值电流密度从(-3.2±1.5)pA.pF-1减少到(-1.6±0.8)pA·pF-1(P<0.01)和(-1.4±0.7)pA·pF-1(P<0.01);但对ICa-L激活动力学曲线无明显影响。结论NP可剂量依赖性抑制豚鼠心室肌细胞ICa-L。

Abstract:Objective To observe the effects of nonylphenol(NP)on L-type Ca2+       currents(ICa-L)in isolated guinea-pig ventricular myocytes.Methods The       ventricular myocytes were isolated from guinea-pig hearts.The L-type Ca2+       currents in the ventricular myocytes treated with NP were measured with       whole cell patch-clamp technique.Results Nonylphenol inhibited ICa-L at       different potentials.Nonylphenol(10-6 mol·L-1,10-5 mol·L-1)reduced the       peak amplitude of ICa-L from-3.2±1.5 pA·pF-1 to-1.6±0.8 pA·pF-1(P <       0.01)and-1.4±0.7 pA·pF-1(P < 0.01),respectively.Nonylphenol did not       influence the activation curve of ICa-L significantly.Conclusion       Nonylphenol could inhibit the L-type calcium currents in isolated       guinea-pig ventricular myocytes with a concentration-dependent manner.
