细胞角蛋白20单克隆抗体免疫胶体金试纸条的研制

目的 研制用于尿液检查的细胞角蛋白20单克隆抗体免疫胶体金试纸条(CK20试条),建立一种无创性筛选诊断和检测膀胱癌复发的新的检测手段.方法 将经处理的CK20单抗标记在胶体金颗粒上并活性进行鉴定,合格后吸附在硝酸纤维膜上并固定在长方形硬质塑料胶片一端为测定线;同时将吸附有IgG蛋白胶体金的硝酸纤维膜固定在测定线的一侧为质控线.将制成的CK20试条吸样端浸入尿液测定.结果 成功的制备CK20试条,初步临床用于膀胱癌检测,敏感性78%(39/50),特异性为87%.结论 采用该方法制备的CK20试条对膀胱癌患者尿液的检测具有较好的灵敏度和特异性,可用于膀胱癌瘤患者筛检诊断和复发的检测.

Research of the strip of immunocolloidal-gold with monoclonal antiCK20 antibody

Objective To explore the test strip of immunocolloidal gold with monoclonal antiCK20 antibody in order to screen bladder tumor by examine urine of patents. Methods Colloidal gold-labeled monoclonal antiCK20 antibody was absorbed on nitrate cellulose membrane, and then the membrane was fixed on plastic strip with double face glue as checking line. On the side of checking line, another piece of membrane with colloidal gold-labeled IgG protein was set as control line. Then the strip was used to check urine of patient with bladder tumor and healthy people. Results Positive rate of this method was 78% (39/50) and specificness was 87% in detecting urine of TCC. Conclu-sion The method for screening bladder tumor by examine urine of patents is set up successfully, which possesses better sensitivity and specificity.
