小鼠性决定基因SRY探针的制备

摘    要:目的 研究制备地高辛标记的小鼠性决定基因SRY探针,用于检测小鼠体内SRY基因表达的研究。方法 按已知的雄性小鼠Y染色体上性决定蛋白基因(SRY)的序列,请上海博亚公司合成四条oligoDNA,采用PCR技术连接并扩增,地高辛标记的方法制备基因探针。结果 用细胞原位杂交方法证实这种探针具有较高的敏感性和特异性。结论 小鼠性决定基因SRY探针的制备成功为进一步研究异体雄性小鼠骨髓移植到雌性小鼠体内后的分布和表达提供了实验基础。

Abstract:Objective To prepare Digoxin - labeled cDNA probe of mouse sex - determining gene, SRY in order to detect SRY gene expression in the micle. Methods According to SRY sequence of male mouse Y chromosome, the four oligo - DNA were synthesized by Bioasia Corp. One SRy probe was successfully prepared by connection and PCR amplification and Digoxin- labeling methods.Results Cellular in situ hybridization showed that the sensitivity and specificity of the probe was relatively high. Conclusion The successful preparation of mouse SRY gene probe provides a next experimental foundation for detecting the distribution, expression of SRY gene in female mice receiving transplantation of male mouse bone marrow cells .
