屋尘螨和粉尘螨主要变应原的筛选和分析

目的研究尘螨主要变应原组分及性质。方法用SDS—PAGE、Western印迹和LC—MS／MS方法，对屋尘螨和粉尘螨的主要变应原组分进行筛选、分析和鉴定。结果两种尘螨的三个样本均有数条在蛋白大小和含量方面较为独特的条带，其中屋尘螨和粉尘螨抗原间蛋白质构成差别较大，两个粉尘螨样本间蛋白带差别较小。屋尘螨主要变应原大小约为12～15kDa和94～98kDa；粉尘螨主要变应原大小约为49～53kDa和94～98kDa，但其中一粉尘螨样本在49～53kDa和94～98kDa位置却未出现反应带。用LC—MS／MS方法从主要变应原条带中鉴定出屋尘螨变应原蛋白约64个，粉尘螨变应原蛋白约84个。它们包括一些已知的尘螨变应原、酶类、translation elongation factor、ribosomal protein、外膜（脂）蛋白、Keratin、Glycine cleavage systemprotein、binding protein、Heatshock protein、regulation protein、hypothetical protein、polpolypmtein和pol protein等蛋白质类别。结论屋尘螨和粉尘螨主要变应原构成及其免疫学反应强度均存在一定区别。针对中国人的尘螨主要变应原可能包括Derp2、Derp14、Dert3，Derf15和Derf17等，上述鉴定的其它蛋白质的一些也可能是潜在的变应原。
