金雀异黄素对晚期蛋白氧化产物刺激大鼠系膜细胞转化生长因子β1分泌的影响

摘    要:目的观察金雀异黄素(Gen)对晚期蛋白氧化产物(AOPPs)刺激下SD大鼠肾小球系膜细胞(MC)分泌转化生长因子β1(TGF-β1)的影响,从而了解作为大豆蛋白质主要成分的Gen对糖尿病肾脏病(DKD)系膜细胞的影响,进而探讨大豆蛋白饮食对DKD的治疗意义。方法以体外培养的SD大鼠MC为研究对象,分为正常培养的对照组(C组)、培养液中加入AOPPs的A组和加入10、50、100、200umol/LGen的G(10)、G(50)、G(100))、G(200)组以及培养液中同时加入AOPPs和Gen的A+G(10)、A+G(50)、A+G(100)、A+G(200)组。酶链免疫吸附(ELLSA)法检测各组细胞培养液中TGF-β1的浓度。结果培养后第12h、24h、48h,A组细胞培养液TGF-β1含量较C组升高(P〈0.05或P〈0.01);G(10)、G(50)组与C组无差异(P〉0.05),G(100)、G(200)组较C组升高(P〈0.05或P〈0.01);A+G(50)、A+G(100)、A+G(200)组较A组和G组降低(P〈0.05或P〈0.01)。A+G(50)、A+G(100)、A+G(200)组培养后第24h较培养后第12h升高;而培养后第48h较培养后第24h升高(P〈0.05或P〈0.01)。结论Gen能减少AOPP刺激下系膜细胞TGF-β1的分泌,提示大豆蛋白饮食可能对DKD患者有益。

Effect of Genistein on the expression of transforming growth factor β1 secreted by cultured rat MCs stimulated by advanced oxidation protein products

Abstract:Objective To find out the effects of genistein (Gen) on the secretion of transforming growth factor β1 by cultured SD rat mesangial cells(MCs)stimulated by advanced oxidation protein products (AOPPs),investigate the effects of Gen on the MCs in the diabetic kidney disease (DKD), and further explore the implication of soy protein diet to the DKD patients. Methods The in vitro cultured SD rat MCs were divided into four groups as control group (C),AOPPs group(A),Gen-treated groups(G), and AOPPs combined with Gen group(A + G). The quantity of TGF-β1 in medium was determined by ELISA. Results The results showed that the content of TGF-β1 in group A was higher than in group C (P〈0. 05 or P〈0. 01 ), there was no significant difference between Gen-treated groups(10 and 50 umol/L) and group C(P〈0. 05), but that in Gen-treated groups(100 and 200umol/ L)was higher than in group C(P〈0. 05 or P〈0. 01). In groups A+ G (50umol/L) ,A+ G(100umol/ L),A+ G (200 umol/L) the content of TGF-β1 was lower than in groups A and group G(P〈0. 05 or P〈0. 01). Further more, the contents of TGF-β1 in groups A + G (50 umol/L), A + G (100 umol/ L) ,A+ G(200 umol/L)after culture for 12 h were higher than in 24-h culture groups, those in 48-h culture groups were higher than in 24-h culture groups. Conclusions AOPPs could promote the expression of TGF-β1 in MCs in vitro, and Gen could down-regulated the secretion of TGF-β1 in a dosedependent manner. It is speculated that soy protein diet might be benefit to DKD.
