摘    要:目的 建立从中华眼镜蛇毒 (Najanajaatra)中大批量分离、纯化natrahagin的方法。 方法与结果 10 g眼镜蛇毒经超滤去除小分子组分后 ,再经Heparin -SehparoseCL - 6B亲和层析及SephadexG - 15 0分子筛层析 ,获得 91mgnatrahagin ,产率为0 91% ;SDS -PAGE显示natrahagin呈单一区带 ,相对分子质量约 4710 0 ,并具有水解纤维蛋白原和血小板膜糖蛋白Ib(PlateletmembraneglycoproteinIb ,GPIb)。 结论此实验成功建立的从中华眼镜蛇毒中大批量制备natrahagin的方法具有高效、简便的特点。

Abstract:Objective To establish a method for natrahagin purification in large batch Methods and Results Ten grams of Naja naja atra venom underwent ultrafiltration to remove the components of low molecular weight Then the remnent was applied to Heparin-Sepharose CL-6B column and Sephadex G-150 column successively Purified natrahagin was 91 mg, ie, the yield was 0 91 % SDS-polyacrylamide gel electrophoresis (SDS-PAGE) showed that natrahagin migrated as a single band of 47 100 and that natrahagin degraded A(-chains of fibrionogen and platelet membrane glycoprotein Ib Conclusion The method established in this study is simple and effective for the purification of natrahgin in large batch
