胆固醇检测试剂的校准偏差和基质效应

摘    要:目的 试验胆固醇试剂的校准偏差和基质效应。方法 用酶法和高效液相色谱法测定新鲜血清和不同基质类型的校准注胆固醇浓度,比较两法的测定结果。结果 多数被检试剂-校准物系统存在校准偏差(=3.5%至+9.4%);所有被检试剂对冻干血清、胆固醇水溶液等制备物有不同程序的基质效应,而冰冻人血清基质效应最小。结论 制备胆固醇校准物需充分认识指定度剂条件下的基质效应,根据新鲜血清测定结果,做出正确定值。

Abstract:Objective To test the calibration bias and matrix effects of cholesterol reagents.Methods Cholesterol concentrations of fresh human serum and samples of different matrix type were measured by enzymatic and liquid chromatographic methods. The data obtained by enzymatic methods were compared with those by liquid chromatographic method.Results Calibration bias were observed on some of the tested reagent calibrator systems and most of the tested reagents showed matrix effects toward lyophilized serum materials, cholesterol of water solution and other processed materials.Conclusion Matrix effects under conditions of given reagent need to be clarified and compensated in assigning the target values of cholesterol calibrators.
