补肾中药调节溴隐亭致大鼠流产模型泌乳素释放肽及其受体表达对妊娠结局的影响

摘    要:目的 研究补肾中药对下丘脑泌乳素释放肽 (PrRP)mRNA及垂体、蜕膜、胎盘组织PrRP受体 (PrRP R)mRNA表达的影响。 方法  6 0只SD孕鼠于孕 6~ 8d皮下注射溴隐亭 ,致成大鼠流产模型后随机分为 5组 :A组为模型组 ,B~D组分别为模型加用中药、泌乳素 (PRL)及孕酮 (P) ,E组为正常妊娠。孕 12d处死大鼠 ,逆转录聚合酶链反应 (RT PCR)方法检测下丘脑PrRP及垂体、蜕膜、胎盘组织PrRP RmRNA表达。 结果 B组PrRPmRNA表达明显高于A组 (P <0 .0 5 ) ,与其他 3组则无明显差别 ;A组垂体PrRP RmRNA表达明显高于其他 4组 (P <0 .0 1) ,各组蜕膜及胎盘PrRP RmRNA表达无明显差异。 结论 补肾中药通过调节下丘脑PrRPmRNA及垂体PrRP RmRNA表达调节PRL合成及分泌

Abstract:Objective: To investigate the effect of “Bushen” preparation on the prolactin-releasing peptide (PrRP) mRNA expression in hypothalamus and PrRP-receptor (PrRP-R) mRNA expression in pituitary,decidua and placenta.Methods:Sixty pregnant SD rats were divided into 5 groups randomly. The rats of group A to D were injected with bromocriptine QD subcutaneously at day 6,7,8 of pregnancy to induce abortion. Group A served as an abortion model. Rats in group B were supplemented with "Bushen" preparation intragastrically,and rats in group C and D supplemented with the injection of PRL and progesterone respectively. Rats in group E were normal pregnant rats with no treatments. All rats were sacrificed at day 12 of pregnancy and the expression of PrRP mRNA in hypothalamus,PrRP-R mRNA in pituitary,decidua and placenta was measured by RT-PCR.Results: PrRP mRNA expression in hypothalamus of “Bushen” treated rats was significantly higher than that of abortion models. There was no significant difference of PrRP mRNA expression among group B,C,D and E. In group A,the expression of PrRP-R mRNA in pituitary was greatly higher than any other groups.Conclusion: “Bushen”preparation regulates PRL synthesis to maintain early pregnancy by the modulation of hypothalamus PrRP and pituitary PrRP-R expression.
