外源性表皮生长因子与神经生长因子对鼠坐骨神经再生影响的比较研究

摘    要:目的:研究外源性表皮生长因子(EGF)与神经生长因子(NGR)对鼠坐骨神经再生的影响。方法:雄性SD大鼠72只随机分成3组,即损伤对照组、EGF组、NGF组。分别于术后2周、4周、6周作坐骨神经功能指数(SFI)、潜伏期、诱发电位、组织学检测、电镜超微结构观察。结果:坐骨神经功能指数恢复率各时间点EGF组、NGF组均明显优于对照组,EGF与NGF之间无明显差异。潜伏期延迟率EGF组、NGF组明显好于对照组,EGF组与NGF组无明显差异。诱发恢复率EGF、NGF组明显优于对照组,EGF组与NGF组间无显著差异。组织学检查:有髓神经纤维计数在2周、4周时EGF组、NGF组明显多于对照组,EGF组与NGF组无显著差异。有髓纤维直径及截面积各时间点EGF组与NGF组均明显优于对照组,EGF组与NGF组之间无明显差异。超微结构观察,EGF组与NGF组再生神经有髓纤维数,髓鞘厚度及髓鞘的成熟度明显优于对照组。结论:外源性的EGF与神经生长因子比较均能促进坐骨神经再生,促进神经功能的恢复。

Abstract:To study the influence of exogenous epidermal growth factor (EGF) and nerve growth factor (NGF) on sciatic nerve in rat.Methods:Seventy-two male Sprague-Dawely rats,weight 180~220g ,were randomly divided into three groups.The sciatic functional index (SFI), Lat, MAP, histomorphometric study included the number of regenerating myelinated nerve fiber;the mean diameter of regenerating myelinated nerve fiber;the cross section area of regenerating myelinated nerve fiber were tested 2,4,6 weeks after operation respectively. All value wound be compared with contralateral side excluding sciatic functional index. Ultrastructure was observed.Results:The recovery of sciatic functional index, Lat and MAP were significantly higher in all time point in EGF, NGF groups than that in control groups. There was no significant difference between EGF and NGF groups. The number of regenerating myelinated nerve fiber 2 and 4 weeks after operation in EGF, NGF groups was much higher than that in control groups. The mean diameter and cross section area of regenerating myelinated nerve fiber in EGF and NGF groups were much larger than that in control groups in all the time point. There was significant difference. But there was no difference between EGF and NGF groups.The number of mylinated nerve fiber, thickness and degree of maturity of regenerating myelinated nerve fiber in EGF and NGF group was superior to that in control groups and the number of degenerating nerve fiber in EGF and NGF groups was lower than that in control group by eletromicroscope observation in all time point.Conclusion:Exogenous nerve growth factor (NGF) and epidermal growth factor (EGF) can facilitate the recovery of nerve function.
