血管平滑肌的ATP敏感钾通道

摘    要:ATP敏感钾通道(KATP)是一类较广泛分布的内向整流钾通道。在生理状态及某些病理条件下KATP参与血管张力的调节。KATP活性受多种因素的调控,胞内二磷酸核苷酸(NDPs)、钾通道开放剂(KCOs)等可激活通道,而ATP和硫脲类药物则特异性抑制通道的开放。分子生物学研究证明KATP由Kir60和硫脲类受体(SUR)共同组成,Kir60构成K+可穿透的通道核心,SUR受体构成通道的调节单位。血管平滑肌中的KATP是由Kir61和SUR2B组成的四聚体结构。但两种亚单位如何联结成有功能的多聚体还需进一步的证明。

Abstract:ATP sensitive K channels(K ATP ) are a widely distributed group of inwardly rectifier K channels. K ATP involves in the regulation of vascular tone on the physiological and pathological conditions. The K ATP channels are stimulated by intracyte nucleotide diphosphates (NDPs) and K channels openers, and inhibited by ATP and sulfonylurea compounds. It was confirmed by molecular biology researches that K ATP was composed of Kir 6 0 serving as a pore forming subunit and sulofnylurea receptor (SUR) as a regulatory protein subunit. In vascular smooth muscle the K ATP is a tetrameric complex by Kir6 1 and SUR2B. It remains to be determined how to connect Kir and SUR to be a functional complex.
